Can we just stop and talk about how Castiel leans his head down further to be sure of what Dean is doing? Innocent like a small pup, almost unable to believe that Dean is truly laughing or that he has caused him to laugh. You see a follow up to this reaction with a touch of perhaps, genuine happiness when Dean pats his back. I believe he felt appreciated in that moment and perhaps intrigued that Dean thought he had done something genuinely amusing.
